
One promising application of CRISPR / Cas9 is to create targeted mutations to introduce traits into an organism domesticated flowers. However, the main current limitations for crop and livestock improvement is to identify the right genes and genetic changes that must be engineered to acquire attractive properties. Here, we discuss the advantages of bio-inspired genome editing, namely the introduction of engineered natural mutation that has been linked to traits of interest in the other lineage (descent, populations or species).
To get a landscape view of potential targets for genome editing, we use Gephebase (www.gephebase.org), manually curated database compilation of published data on genes responsible for evolutionary changes and domesticated throughout eukaryotes, and examine> 1200 mutations have been identified in the coding region of more than 700 genes in animals, plants and yeast. We observed that our genetic knowledge is relatively important for certain traits, such as resistance xenobiotics, and poor for others. We also note that protein-null allele, often because of nonsense and frameshift mutations, representing a significant portion of the locus is known domestication (42% of the mutations identified coding), compared to intraspecific (27%) and the evolution of interspecific (11%).
Although this trend may be subject detection, publications and curatorial bias, it is consistent with the notion that the breeders have selected mutations-effects of the underlying properties of adaptive in specific settings, but that these mutations and phenotypes related will not survive the vagaries of external changes and internal environment. Our compilation of evolution and domestication loci reveal interesting options for bio-inspired and free transgene genome editing.

Development of a 3D heart chitosan porous scaffold / gelatin for bioartificial liver device.
functional artificial heart (Velasquez), with embedded hepatocytes which performs the functions of a normal liver, have been developed over the last decade. It is important to note that the liver scaffold, which is a biological core functional bioartificial liver plays an important role in the bio-cartridge in a bioartificial liver.
In this study, three-dimensional (3D) scaffold for the in vitro culture of liver were made by freeze-drying chitosan / gelatin (CG) solutions. A CG scaffold has advantages such as (i) the cost and easy-to-make; (Ii) easy to fabricate with different modulus press by changing the concentration of glutaraldehyde; (Iii) non-cytotoxicity; and (iv) the porous structure similar to the extracellular matrix (ECM), thus facilitating the adhesion and proliferation of hepatocytes. The results showed that the modulus press and maintainability of scaffolding CG correlated with increased glutaraldehyde.
Furthermore, hepatocyte survival and liver function showed the best performance with 0.61% glutaraldehyde-CG scaffolding. This CG scaffold not only have a higher hepatocyte biocompatibility and mechanical strength, but also preserve liver function and viability in vitro culture; particularly, the mechanical properties of 0.61% glutaraldehyde-CG scaffolding that is very similar to that in the normal liver. CG scaffolding as liver scaffold may have a high potential for further bioartificial liver designs in the near future.

Description: Gentaur Mononuclear Cells (MNC) represent the enriched lymphocyte and monocyte fraction of whole blood. They are isolated from umbilical cord blood and adult peripheral blood of healthy donors at Gentaur's cell culture facility. Mononuclear Cells from cord blood contain a relatively high percentage of primitive progenitor cells,whereas Mononuclear Cells from adult peripheral blood (hMNC-PB) contain large numbers of mature immune cells.Note: When culturing pooled Peripheral Blood Mononuclear Cells, a bidirectional allogeneic mixed lymphocyte reaction (MLR) is likely to occur during the first 48 hours. For HIV-related research, prior CD8+ T-cell depletion is recommended.Freshly collected whole blood is carefully separated by optimized low-density gradient centrifugation, effectively removing the granulocytes.
