AAV-DJ ZsGreen |
||
BPS Bioscience | 500 µl x 2 | EUR 595 |
Description: Adeno-Associated Virus-DJ (AAV-DJ) is a synthetic serotype made from eight different wild-type AAV serotypes (AAV2, 4, 5, 8, 9, avian, bovine, and goat AAV) using DNA shuffling. These modifications give the AAV-DJ serotype improved transduction efficiency in vitro and in vivo compared to wild-type serotypes. Consequently, AAV-DJ can infect a broad range of cell types._x000D_These AAV particles constitutively express ZsGreen under a CMV promoter. ZsGreen is a human codon-optimized variant of the green fluorescent protein isolated from reef coral (Zoanthus sp). It has been engineered for higher expression in mammalian cells and is up to four times brighter than enhanced GFP (eGFP). ZsGreen expression and AAV transduction efficiency can easily be verified and optimized by fluorescence microscopy or flow cytometry. ZsGreen has an excitation wavelength of 493 nm and an emission wavelength of 505 nm. |

F49626-0.08ML | NSJ Bioreagents | 0.08 ml | EUR 140.25 |
Description: Park7/DJ-1 acts as positive regulator of androgen receptor-dependent transcription, and may function as redox-sensitive chaperone and as sensor for oxidative stress, as well as preventing aggregation of SNCA. This protein has been shown to protect neurons against oxidative stress and cell death, and to play a role in fertilization. It is detected in tau inclusions in brains from neurodegenerative disease patients, and is generally highly expressed in pancreas, kidney, skeletal muscle, liver, testis and heart, with detectable levels in placenta, brain, astrocytes, Sertoli cells, spermatogonia, spermatids and spermatozoa. Defects in Park7/DJ-1 are the cause of autosomal recessive early-onset Parkinson disease 7, a form of Parkinson disease characterized by onset before 40 years, slow progression and initial good response to levodopa. |

Description: Parkinson disease 7, also known as Protein deglycase DJ-1, is a protein which in humans is encoded by the PARK7 gene. PARK7 belongs to the peptidase C56 family of proteins. It acts as a positive regulator of androgen receptor-dependent transcription. It is also involved in tumorigenesis and in maintaining mitochondrial homeostasis. DJ-1 may function as a redox-sensitive chaperone, as a sensor foroxidative stress, and it apparently protects neurons against oxidative stress and cell death. It has been found that DJ-1 mutations that impair transcriptional coactivator function can render dopaminergic neurons vulnerable to apoptosis and may contribute to the pathogenesis of Parkinson disease. |

These modifications give the AAV-DJ serotype improved transduction efficiency in vitro and in vivo compared to wild-type serotypes. Consequently, AAV-DJ can infect a broad range of cell types. _x000D_These AAV particles constitutively express the firefly (Photinus pyralis) luciferase and enhanced Green Fluorescent Protein (eGFP) genes connected via a T2A linker, under the control of a CMV promoter. The T2A self-cleaving peptide (derived from Thosea asigna virus 2A) leads to the efficient cleavage of the transcript, and expression of luciferase and eGFP as two separate proteins. |
